5 DPO: The Blastocyst Stage and the Urge to Test
A big milestone for the embryo. Still nothing for a test to find.

Five days past ovulation is often when the temptation to test gets strong. You are a third of the way through the wait, you may feel different, and there is a box of tests in the cupboard. This page explains why a test at 5 DPO cannot help yet, what a "test early" claim on the box really means, and what is happening while you wait.
What happens at 5 DPO
If an egg was fertilized, day 5 is when it becomes a blastocyst: a fluid-filled structure with an inner cell mass that will become the embryo and an outer layer, the trophoblast, that will help form the placenta.1 It is in the uterus but still enclosed in its protective shell. Tomorrow, around day 6, it hatches out of that shell and becomes ready to implant.1
Your progesterone is now close to its peak, which comes 6 to 8 days after ovulation.2 That timing matters for symptoms: the days around 5 to 8 DPO are often when people feel the most "pregnant", in every cycle.
5 DPO symptoms
Breast soreness, bloating, tiredness and moodiness at 5 DPO are explained by your luteal-phase hormones, not by a blastocyst that has not yet touched the uterine lining. Feeling exactly as you did before past periods is normal, and so is feeling different. Neither predicts the outcome.
Can you get a positive pregnancy test at 5 DPO?
No. hCG is made only once an embryo starts implanting. In the NIH study of 221 women, hCG first appeared 6 to 12 days after ovulation in continuing pregnancies, and 84% implanted on day 8, 9 or 10.5
Daily urine data show how little hCG there is even a few days later. A study of 109 women who conceived measured urine hCG each day after ovulation:3
| Days past ovulation | Median urine hCG | 90% of pregnancies were below |
|---|---|---|
| 5 | No data; before implantation | n/a |
| 7 | 0.00 mIU/mL | 0.20 mIU/mL |
| 8 | 0.06 mIU/mL | 2.91 mIU/mL |
| 9 | 4.04 mIU/mL | 11.32 mIU/mL |
In a 2011 lab comparison, the most sensitive home tests detected about 5.5 mIU/mL, while others needed about 22 mIU/mL.8 At 5 DPO, the numbers are not even on the chart.
What "6 days sooner" on the box means
First Response says its Early Result test detects hCG "as early as 6 days before your missed period", which it defines as 5 days before the day your period is expected.4 That counts back from your period, not forward from ovulation. In a large app-based dataset, the average luteal phase was 12.4 days, and most fell between 7 and 17 days.7 So "5 days before your expected period" lands around 7 DPO for someone with a 12-day luteal phase and 9 DPO for someone with 14 days. It never means 5 DPO.
5 DPO and implantation
Implantation has not started. The blastocyst must hatch first, around day 6.1 If you notice spotting now, it is too early to be implantation bleeding. Our guide on when implantation bleeding occurs covers the real window.
What to do now
Put the tests away for now. The FDA notes that testing too early can give a false negative, and that the most reliable results come after a missed period.6 An early negative can feel crushing even though it means nothing yet.
Plan when you will test. The DPO calculator gives you a date, and our guide on how many days after ovulation you can test explains why. If you want to test before your period, choose a sensitive test.
Get support for the middle of the wait. This is often the hardest stretch. Our page on coping with the two-week wait can help, and we can send you short reminders timed to your cycle.
Next: 6 DPO, the earliest day implantation has been recorded. Not pregnant this cycle? Track smarter next time.
